Technical Description

EX-PRESS 293™-BL is a derivative of EX-PRESS 293™, a HEK293 cell line adapted to be maintained in chemically-defined serum-free media and suspension culture, with the stable integration of biotin ligase for in cellulo biotinylation. EX-PRESS 293™-BL cells have been optimized for high density, transfection efficiency, and recombinant biotinylated protein production using a protocol designed to reduce transfection requirements without sacrificing recombinant protein production or quality.

Expressions of recombinant proteins with an Avi-Tag™ are efficiently biotinylated for downstream applications when production media is supplemented with exogenous biotin. The TGEX™ mammalian expression vectors vectors are recommended for recombinant antibody production.

Frozen cells are available as a single cryovial, or as a multipack to eliminate the need to expand and bank cells before use. Each cryovial contains 1 ml of 1 x 107 cells/ml in BalanCD HEK293 with 10% DMSO. Cells should be thawed directly in complete BalanCD HEK293 supplemented with 2-mM L-glutamine.

Thawed cells are grown in a humidified CO2 incubator on a shaking platform in multi-well plates to large shake flasks. Additionally, EX-PRESS 293™ cells take advantage of the unique shape of Thomson Optimum Growth® shake flasks in transfecting cells at high density followed by increasing production volume without needing to change vessels. This allows for increased yields at lower cost and shaker footprint, without impacting cellular growth. Recombinant protein production follows a simple fed-batch protocol with harvest six days post-transfection.

Notes

Thawing EX-PRESS 293™-BL cells:

To ensure the highest level of viability, thaw a cryovial and initiate culture upon receipt. If immediate culturing of cells is not required, transfer frozen cryovial(s) to storage in liquid nitrogen. Cryovials stored at -70°C will result in loss of viability.

Transient transfection and protein production:

EX-PRESS 293™-BL cell lines are compatible with commercially available transient transfection systems and mammalian expression vectors. Protocols for transfection and a short 6-day fed-batch recombinant protein production are available in the product manual.
