EBY100 Yeast Cells
Technical Description
Saccharomyces cerevisiae EBY100 is a specialized MATα yeast strain engineered for high-efficiency surface display and protein screening. Derived from the protease-deficient BJ5465 background, this strain is designed to maintain the structural integrity of displayed proteins, making it an ideal host for directed evolution and antibody library maturation. EBY100 features a genomic integration of the AGA1 anchoring protein under the galactose-inducible GAL1 promoter, ensuring that the display machinery is only activated when desired. The system operates using the natural α-agglutinin yeast adhesion mechanism. While the EBY100 host cell provides the cell-wall-bound Aga1p subunit, the user-supplied vector (e.g., pCTCont2, pYD1 or pYDL series) expresses the target protein fused to the Aga2p subunit. Once induced in galactose-supplemented media, these two subunits link via disulfide bonds, effectively tethering the target protein to the extracellular matrix. With leu2 and trp1 selection markers and a robust growth profile, EBY100 provides a reliable, high-density display platform compatible with both magnetic bead enrichment and FACS-based screening.
